Im thinking about running spray on my truck.I was told that you could not run NOS on a H.O motor, is this true

No, it's not true.

You can run NOS, Edlebrock, Zex or NX dry nitrous systems on an HO, or you could buy a nitrous system from another company or cobble your own together. They all have great results if you have a lot of fuel down low.

Would i have to get rid of my water/meth to run NOS

Nope. You wouldn't have to get rid of it to run Nitrous Express or Zex either.

However, nitrous and water/meth both result in cylinder pressure increases. I don't know what kind of timing your boxes are giving you, or how much boost your seeing, but when you start putting lots of fumigants into your engine headgaskets tend to start poppin'.

As long as you watch your pressures you'll be fine.

Already seeing 56psi i need studs and o rings bad

Nitrous would probably put you over the edge then. Or, it would probably put you farther over the edge... I guess it wouldn't hurt anything if you had it programmed to only shoot off the line...

But I think it'd be putting the money towards some gasket modification before nitrous with those numbers.
